The ETS (E26-transformation specific) domain transcription factor family that have regulatory roles in numbers of cellular genes as activators or repressors effects fundamental circumstances such as cellular differentiation, proliferation or development etc. These transcription factors categorized into several members as their sequence homologies contain common purine-rich DNA binding sequence in all members. Pea3 (ETV4/E1AF) transcription factor which is one of the ETS family member known to be regulated by specific growth factors through MAPK/ERK pathway by phosphorylation of Serine/Threonine residues during neural development. In the thesis, we fundamentally focused the question of which growth factor regulates which phosphorylation sites on Pea3. Substitutional mutation on putative phosphorylation sites was performed to understand whether these motifs are indeed important in growth factor induced activity of Pea3. Firstly, putative phosphorylation sites of Pea3 were mutated with replacement of amino acids from Serine to Alanine or Glutamic acid. After the transfection of mutant Pea3 plasmids into SH-SY5Y human neuroblastoma cells, various growth factors were applied. The protein expression levels were observed and compared with wild type Pea3 and among themselves by cell culture techniques, immunoprecipitation technique, Western Blot Analysis and SDS-PAGE electrophoresis. Regulation between mutant Pea3 and different promoters was also investigated with luciferase reporter assay as well as Pea3 protein expression studies. This thesis could be provided to help understanding of what kind of effects of growth factors have on Pea3 and to identify Pea3 phosphorylation sites. Pea3 is known to have a role in neurite outgrowth, and the elucidation of Pea3 activation would contribute to understanding of process of neurite outgrowth. Thereby, it would illuminate mechanisms of neuroregeneration.
