Widespread use of fungicides in both clinical and agricultural areas has caused the fungi to gain tolerance. Growing concern over this issue has led to the search for alternative approaches. In this context, photodynamic inactivation (PDI) is a promising alternative to conventional fungicides. PDI basically consists of three basic components: light, oxygen, and photosensitizer (PS). In this thesis, the properties of two different dye (BODIPY and Rose Bengal) as photosensitizers in PDI applications were evaluated on conidia of maize (Zea mays)-pathogenic fungus species Fusarium verticillioides. The efficacy of PDI with each PS was determined by the comparative minimum inhibitory concentration (MIC) and the survival of the conidia number under and without light. The amount of intercellular reactive oxygen species (ROS) was measured in a fluorometric assay using 2',7'-dichlorofluorescein diacetate staining on PS-treated F. verticillioides conidia (spores). The effects of pathogenicity of PSs-treated spores on maize leaves were also investigated. At the molecular level, the expression of Sterol 14 a-demethylase (Cyp51), mitochondrial cytochrome b (Cytb) and succinate dehydrogenase (SDH) genes in response to PS application were determined by qRT-PCR. As a result, it was observed that the degree of inhibition of fungal growth increased with PS concentration. The increased production of ROS due to PS application suggests that photoactivated reactive oxygen by PSs induces a local cell death that inhibits fungal growth and pathogenicity in leaves. These findings present PSs as a new fungicide for PDI.
