Optimization of 2,3-Butanediol production in Streptomyces coelicolor

Streptomyces'ler gram pozitif toprak bakterileridir ve esas olarak tıbbi biyoteknolojide kullanılan ikincil metabolitleri ürettiği bilinmektedir. Sekonder metabolit üretme yeteneklerinden dolayı Streptomyces üzerine yapılan araştırmaların çoğu bu konuya odaklanmıştır. Bildiğimiz kadarıyla literatürde Streptomyces'in primer metabolizmasına odaklanan çok az çalışma bulunmaktadır. Doğal olarak üretilebilen ve birincil karbon metabolizmasına bağlı olan 2,3-Butandiol (2,3-BDO), sentetik plastikleştiriciler, kauçuklar, fumigantlar gibi farklı türdeki endüstriyel ürünlerin üretiminde kullanılan bir glikol alkoldür. Ayrıca yakıt katkısı, oktan arttırıcı ve antifriz maddesi olarak da kullanıldığı bilinmektedir. Plastik, aroma, kozmetik ve ilaç endüstrilerinde hammadde olarak kullanılması 2,3-BDO'yu çok değerli kılmaktadır. Laboratuvarımızda yapılan önceki çalışmalarda Streptomyces coelicolor'un 2,3-BDO üretebildiği kanıtlanmış ancak üretim miktarı oldukça düşük bulunmuştur. Bu çalışmanın amacı, hem yabanil tip S. coelicolor suşunun hem de glikoz taşıyıcı proteinin üretiminden sorumlu olan glcP1 geninin ekstra bir kopyasını içeren recombinant S. coelicolor suşunun 2,3-BDO üretimini optimize etmektir. 2,3-BDO üretimini arttırmak için farklı besiyerleri (YEME, TSB ve TSB+10% glukoz), pH (pH 5.0, pH 6.0, pH 7.0, pH 8.0 ve pH 9.0) ve sıcaklıklar (25°C, 30°C ve 35°C) test edildi. HPLC sonuçlarına göre, hem yabanil tip hem de rekombinant Streptomyces suşları tarafından 2,3-BDO'nun üretilmesi için en uygun ortamın ve koşulların, pH 7.0 ve 30°C sıcaklıkta 10% glukoz içeren TSB olduğu saptanmıştır. Bu koşullarda üretilen maksimum 2,3-BDO miktarı yabanil tip S. coelicolor için 0,033 mg/mL, rekombinant S. coelicolor için ise 0,035 mg/mL olarak bulunmuştur. Endüstriyel proseslere göre değerler şu anda düşük olsa da gelecekte yapılacak optimizasyon ve genetik çalışmalarla daha yüksek üretim seviyelerine ulaşılabilir. Bu tez, Streptomyces'in birincil metabolit üretimi için ön veriler sağlaması açısından önemlidir.
Streptomyces are gram-positive soil bacteria and are known for producing secondary metabolites mainly used in medical biotechnology. Most research on Streptomyces has focused on this subject due to their ability to produce secondary metabolites. To the best of our knowledge there are few studies that have focused on the primary metabolism of Streptomyces in the literature. 2,3-Butanediol (2,3-BDO) is a glycol alcohol that can be produced naturally and is dependent on primary carbon metabolism. It is used in the production of various industrial products, such as synthetic plasticizers, rubbers, fumigants, and as a fuel additive, octane booster and antifreeze agent. Its value lies in its use as a raw material in the plastic, flavor, cosmetic, and drug industries. Previous studies conducted in our laboratory have shown that Streptomyces coelicolor is able to produce 2,3-BDO, but the production amount was found to be quite low. This study aims to optimize the production of 2,3-BDO in both the wild-type S. coelicolor strain and a recombinant strain containing an additional copy of the glcP1 gene responsible for glucose transporter protein production in S. coelicolor. To enhance 2,3-BDO production, different kinds of media (YEME, TSB, and TSB+10% glucose) and growth conditions, including pH (pH 5.0, pH 6.0, pH 7.0, pH 8.0 and pH 9.0) and temperatures (25°C, 30°C and 35 °C) were tested. The HPLC results indicated that the optimal medium and conditions for producing 2,3-BDO by both wild-type and recombinant Streptomyces strains are TSB supplemented with 10% glucose, at a pH of 7.0 and a temperature of 30°C. The maximum amount of 2,3-BDO that could be produced in these conditions was found to be 0.033 mg/mL for S. coelicolor and 0.035 mg/mL for recombinant S. coelicolor. Although the values for industrial processes are currently low, higher production levels can be achieved through future optimization and genetic studies. This thesis is significant as it provides preliminary data supporting the use of Streptomyces for primary metabolite production.
