A biochemical oxygen demand (BOD) sensor, based on an immobilized Pseudomonas syringae in combination with a dissolved oxygen electrode, has been developed for the purpose of analysis of organic compounds in aqueous samples. Microorganisms were immobilized in highly porous micro-cellular polymer disk constituted of oil and water phases by recycling the suspended culture through the reactor for 24 h at a flow rate of 1 ml/min. The sensor measurements were carried out by dissolved oxygen probe placed on the polymer disk using continuous wastewater feeding and flow injection systems. The sensor performance achieved in two measurement systems showed a detection linearity over the range of 5?100 mg BOD5?/L (r2>0.98). While the optimum flow rate was 1.0 ml/min in continuous substrate feeding technique, it was found to be 0.6 ml/min in flow injection technique. Typical response times of the BOD sensor were 60?150 sec. in continuous feeding system and 25?40 sec in flow injection system. The optimal sensor response was observed at pH 6.8, 22 0C and 2.0 mm disk height for both two measurement techniques. For municipal and industrial wastewaters, BOD values estimated by this biosensor correlate well with those determined by the conventional 5?day BOD test.
