İmmobilize glukomilaz ile maltodekstrinden glukoz üretimi

Abstract

Endüstriyel bir enzim olan Glukoamilaz nişastadan glukoz elde etmede kullanılır. Bu çalışmada Aspergillus niger mikroorganizmasından üretilen Glukoamilaz enzimi için yeni bir immobilizasyon metodu geliştirilerek maltodekstrinden glukoz elde edilmiştir. Glukoamilaz ilkönce jelatinleştirilmiş mısır nişastası üzerine adsorblandı. Daha sonra glycerol içerisinde çözülmüş sodyum alginate ile karıştırıldı. Bir enjektör yardımı ile %1 glutaraldehid içeren %1'lik CaCİ2 çözeltisine ilave edilerek fiberler elde edildi. İmmobilizasyon verimi %22 olarak tespit edildi. 20 reaksiyon ve 21 gün boyunca immobilize Glukoamilaz'ın aktivitesinde azalma gözlenmedi.

Glucoamylase, an industrial enzyme, hydrolyzes a- 1,4 and a- 1,6 glucosidic bonds of starch from non-reducing ends to produce glucose. In this study, a novel method for the immobilization of Aspergillus niger Glucoamylase was developed and used to produce glucose from maltodextrin. Glucoamylase first was adsorbed on gelatinized corn starch and mixed with sodium alginate dissolved in glycerol and this mixture was added through a syringe to 1% calcium chloride solution containing 1% glutaraldehyde to obtain alginate fibers. Immobilization yield was 22% and there was no reduction in the activity after 20 batches and within 21 days.
