Nitrite biosensors are studied as an alternative method to measure the nitrite concentrations in situe due to their advantages of measuring quickly, accurately and easily. In this study, a novel apmerometric nitrite biosensor was developed by immobilizing nitrite reductase B (NirB) of Escherichia coli by cross-linking on electrochemically polymerized polyglutaraldehyde/polypyrrole (PGA/PPy) film on the glassy carbon electrode surface. Electropolymerization was carried out with a cell containing pyrrole and polyglutaraldehyde by cyclic voltammetry between -1V and 1.5V (vs. Ag/AgCl) at scan rate 0.05Vs-1. Measurements were carried out in a 0.05 M phosphate buffer solutionThe response time (t95%) of the biosensor was about 10 s and sensitivity was 77.81 mA M-1 cm-2 with linear response range of 1–3500 ?M (R2=0.9935) and detection limit calculated as 2.7 ?M. E. coli Nitrite reductase NirB enzyme, which was previously cloned and characterized by our team, was used for the first time in the preparation of an amperometric nitrite biosensor in this study. This newly purified NirB enzyme has the potential to be used in the development of nitrite biosensors.
