Yapılan tez çalışmasında, metanol ve su çözücüleri kullanılarak elde edilen fındık yeşil kabuğu ekstraktlarının (FM ve FS) fitokimyasal içerikleri ve Bacillus cereus ATCC 11778, Bacillus subtilis ATCC 6633, Streptococcus uberis ATCC 700407 ve Enterecoccus faecalis ATCC 29212 suşları üzerinde antibakteriyel etkileri ile FM ekstraktının antibiyofilm ve anti quorum-algılama etkinliği incelendi. Ekstraktlar % 80 metanol veya saf su ile 50°C sıcaklıkta 30 dakika uygulama süresinde hazırlandı. Ekstraktların toplam fenolik içerikleri Folin-Ciocalteu yöntemi, toplam flavonoid içerikleri alüminyum klorür yöntemi ve antioksidan etkinlikleri DPPH radikali süpürme aktivitelerinin tayin edilmesi ile belirlendi. Ekstraktların antimikrobiyal etkinliğinin incelenmesinde agar kuyu difüzyonu ve MİK yöntemleri uygulandı. FM ekstraktının E. faecalis suşunun 24 ve 48 saatlik biyofilm oluşumlarını ortadan kaldırma etkinliği ve anti quorum-algılama aktivitesi Chromobacterium violaceum bakterisi ile test edildi. FM ekstraktının toplam fenolik (FM: 17.8±1.7, FS: 11.9±1.4 mg GAE g-1), toplam flavonoid (FM: 0.4±0.1, FS: 0.2±0.1 mg QE g-1) içeriklerinin ve DPPH radikali süpürme etkinliğinin (FM: IC50=0.75 mg mL-1, FS: IC50=1.61 mg mL-1) FS ekstraktından daha yüksek olduğu tespit edildi. FS ekstraktının B. cereus, S. uberis, E. faecalis bakterileri, FM ekstraktının ise B. cereus, E. faacelis bakterileri üzerinde gentamisine benzer veya daha fazla etki gösterdikleri görüldü. FM ekstraktının E. faecalis bakterisi üzerinde yüksek antimikrobiyal etkisi (MİK50=0.81 mg mL-1) olduğu ve E. faecalis bakterisinin 24 ve 48 saatlik biyofilmlerine 5 dakika süre ile uygulamasının (1:8 dilüsyonda, 18.4 mg mL-1) % 42 oranında biyofilm oluşumunu ortadan kaldırdığı tespit edildi. Ayrıca, FM ekstraktının anti quorum-algılama etkinliği gösterdiği saptandı. Bu çalışmanın sonuçları, fındık yeşil kabuk esktraktlarının gıda, kozmetik veya ilaç endüstrileri için potansiyel doğal antioksidan ve antimikrobiyal ajan kaynakları olabileceği belirlendi.
In the thesis study, the phytochemical contents and antibacterial effects of hazelnut green leafy cover extracts (FM and FS) obtained by using methanol and water were assessed. The antibacterial effect of extracts were evaluated against Bacillus cereus ATCC 11778, Bacillus subtilis ATCC 6633 Streptococcus uberis ATCC 700407 and Enterecoccus faecalis ATCC 29212. The antibiofilm and anti quorum sensing efficacy of FM extract was also determined. Extracts were prepared with 80% methanol or pure water at 50 °C for 30 minutesThe total phenolic contents of the extracts were evaluated by the Folin-Ciocalteu method, the total flavonoid contents were evaluated by the aluminum chloride method, and the antioxidant activity was evaluated by determining the DPPH radical scavenging activities. Agar well diffusion and broth dilution methods were used to investigate the antimicrobial activity of the extracts. The biofilm removal effect of the FM extract tested against E. faecalis. Anti quorum-sensing activity was tested with Chromobacterium violaceum. Total phenolic (FM: 17.8±1.7, FS: 11.9±1.4 mg GAE g-1) and total flavonoid (FM: 0.4±0.1, FS: 0.2±0.1 mg QE g-1) contents and the DPPH free radical scavenging activities (FM: IC50=0.75 mg mL-1, FS: IC50=1.61 mg mL-1) of FM extract were found to be higher than those obtained by FS extract. It was observed that the FS extract had similar or more effects than gentamicin against B. cereus, S. uberis, E. faecalis, and the methanol extract against B. cereus, E. faecalis. FM extract had high antimicrobial effect (MIC50=0.81 mg mL-1) against E. faecalis. It was determined that applying the extract for 5 minutes (1:8 dilution) to the 24 and 48-hour biofilms of E. faecalis removed the biofilm at a rate of 42 %. It was determined that the FM extract showed anti-quorum-sensing activity. The results of this study provided evidence that hazelnut green leafy cover extracts could be potential sources of natural antioxidant and antimicrobial agents for the food, cosmetic, or pharmaceutical industries.
