Güçlü afiniteye sahip moleküllerin farmakokinetiği in silico ADME ile incelendi. Bu çalışma, P09 molekülünün CRY1'in stabilitesini azalttığını göstermiştir.
Circadian rhythms are biological patterns of approximately 24 hours in biological processes or systems. It is assumed that about 10% of transcriptome or 20-40% of the proteome shows circadian rhythmic patterns. The oscillation at RNA and protein levels indicates that the timing of treatment with drugs is important. Because of this relationship, it is possible to change the efficiency of the drugs by modulating circadian rhythms using small molecules. With the developments in computer-aided drug design in recent years, there has been an increase in the speed of reaching pre-clinical studies. In our study, using the crystal structure of the CRY1-PER2 complex, the interaction side of these proteins was chosen as the target, and molecular docking for, ~2 million molecules were performed. The pharmacokinetics of molecules with strong affinity were examined via in silico ADME. In vitro studies of 14 molecules were carried out by subjecting suitable molecules to molecular dynamics simulation and relative free energy calculation. The sub-toxic dose was determined by MTT. Molecules affecting circadian rhythm were determined by real-time bioluminescence assay. The effect of these molecules on CRY1 was investigated by Western blot. We report that the P09 molecule reduces the stability of CRY1.
