Streptomycetes are Gram-positive, filamentous soil-dwelling bacteria with high G-C content. Recent studies indicated that they have a high potential to produce more variety of secondary metabolites thanks to their cryptic biosynthetic gene clusters in their genomes. A previous study carried out in our laboratory showed that the deletion of polyphosphate kinase gene (ppk), responsible for polyphosphate biosynthesis, increased the production of actinorhodin in both Streptomyces coelicolor strains. So, the deletion of the ppk gene created S. coelicolor mutant strain with a strong endogenous antibiotic production capacity. In the scope of the present study, we investigated whether the S. coelicolor ?ppk strain could also produce primary metabolites in high amounts or be a possible prototype as a heterologous expression host. For that reason, we have selected cholesterol oxidase to determine the effect of ppk deletion on primary metabolism and the 8-demethyl-tetracenomycin (8-DMTC), an anti-tumor agent, to study the potential of ?ppk strain as a host for the heterologous expression. We have successfully cloned the putative cholesterol oxidase gene of S. coelicolor, but the effect of polyphosphate deficiency on enzyme production couldn't be determined because of the lack of substrate (cholesterol) that must be supplemented into the medium. In the case of heterologous expression experiments, the 8-DMTC expressing recombinant S. coelicolor ?ppk strain produced slightly more 8-DMTC than the recombinant wild type. Besides, we have noticed morphological changes in the colony formation of the recombinants caused by the production of 8-DMTC. The recombinant S. coelicolor wild type and ?ppk strains can not form colonies, pellets or clumps. Moreover, they can not produce pigmented antibiotics and do not sporulate, unlike wild type and ?ppk. Our study provided valuable information to the literature since the interesting morphological changes of the S. coelicolor colonies caused by the expression of 8-DMTC have not been reported before.
