Kurşun-çinko maden cevherleri ve atıklarından elde edilen izolatlarla ekonomik elementlerin biyoremediasyonu

Özet
Biyoliçing, mikroorganizmaların metalleri cevherler ya da atıklardan ayrıştırdığı, sürdürülebilir, yüksek enerji harcaması olmayan, uygun maliyetli ve yüksek miktarda metalin liçing işlemi ile elde edilebildiği alternatif bir yöntemdir. Biyoliçing ile metaller ayrıştırılırken aynı zamanda toksik ağır metallerin biyoremediasyonu da gerçekleştirilmiş olur. Bu çalışmada, Balıkesir (Balya) Kurşun-Çinko maden sahasından temin edilen cevher ve flotasyon zuhurları, yerel izolatlar ile biyoliç edilmiştir. Çalışmanın başında cevher ve flotasyon atığının elementel kompozisyonu XRD analizi ile belirlenmiştir. Maden sahasından getirilen cevher ve flotasyon atığı örneklerinden bakteri izolasyonu Tryptic Soy Broth, Tryptic Soy Agar ve Nutrient Agar kullanılarak, uygun sıcaklık ve inkübasyon süresinde yapılmıştır. Biyoliçing çalışması için, 96-120 saat süresince, %10 (w/v) cevher veya flotasyon atığı, Nutrient Broth içerisinde ve aerobik koşullarda 30oC ve 150 rpm'de inkübe edilerek uygulanmıştır. Ön kültürler ve bakteri izolasyonunu takiben 50 ml nispeten büyük ölçekli çalışma yapılmıştır. Bu çalışmada örneklerden 24h, 48h ve 120h saatlerde 10 ml örnekler alınmış, kalan miktar 2 hafta boyunca oda sıcaklığında tutulmuştur. Buna göre; cevher ve flotasyon atığı içeren erlenlerden, üst sıvı ve katı madde ayrılması 3500 rpm'de, 5 dakika boyunca santrifüjlenerek yapılmıştır. Üst sıvılar ayrılmış, kalan çökeltiler de etüvde kurutulmuştur. Biyoliçing aktivitesi için ayrılan üst sıvılar ICP-OES analizi ile, çökelmiş katı maddeler ise XRF analizi ile incelenmiş ve alınan ölçüm sonuçları irdelenmiştir. Çalışmanın sonucunda cevher ve flotasyon atığında bulunan kurşun, çinko ve bakır ve bunlar gibi ağır metallerin, bir adet cevher izolatı (C2) ve bir adet flotasyon atığı izolatı (F8) tarafından farklı oranlarda biyolojik iyileştirme veya biyoliçing aktivitesi gösterdiği belirlenmiştir.
Bioleaching is an alternative method where microorganisms extract metals from ores or waste. Leaching is a sustainable, energy-efficient, and cost-effective method that allows for the extraction of large quantities of metals. While bioleaching separates metals, it also facilitates the bioremediation of toxic heavy metals. In this study, ore and flotation waste obtained from the Balıkesir (Balya) Lead-Zinc Mine were bioleached using local isolates. At the beginning of the study, the elemental composition of the ore and flotation waste was determined by XRD analysis. Bacterial isolation from ore and flotation waste samples from the mine site was performed using Tryptic Soy Broth, Tryptic Soy Agar, and Nutrient Agar at the appropriate temperature and incubation time. For the bioleaching study, 10% (w/v) ore or flotation waste was incubated in Nutrient Broth at 30°C and 150 rpm under aerobic conditions for 96-120 hours. Following pre-cultures and bacterial isolation, a relatively large-scale 50 ml study was conducted. In this study, 10 ml samples were taken at 24, 48, and 120 hours, and the remaining amount was kept at room temperature for 2 weeks. Accordingly, supernatant and solid separation was achieved by centrifuging the flasks containing ore and flotation waste at 3500 rpm for 5 minutes. The supernatants were separated, and the remaining sediments were dried in an oven. The supernatants separated for bioleaching activity were analyzed using ICP-OES analysis, and the precipitated solids were analyzed using XRF analysis, and the obtained measurement results were analyzed. As a result of the study, it was determined that lead, zinc and copper and similar heavy metals in the ore and flotation waste exhibited bioremediation or bioleaching activity at different rates by one ore isolate (C2) and one flotation waste isolate (F8).
