Angiotensin receptors can interact with each other or with other receptors to form GPCR homo- or heterodimers. As a result of dimerization, their signaling pathways or the types of G proteins with which they couple and signal that they generate may change, and their signaling may increase or decrease. In order to elucidate this allosteric mechanism, it is important to determine the interaction interfaces. In this thesis, firstly, the possible homodimer forming interfaces of angiotensin II type 1 receptor (AT1R) were determined. Then, the interaction interfaces of AT1R-AT2R heterodimer formed by AT1R with angiotensin II type 2 (AT2R) were investigated. Interfaces were investigated using molecular dynamics (MD) simulations. Four possible interfaces are proposed for the AT1R homodimer. These are TM1,2,8, TM5, TM4, and TM4,5 tight interfaces, respectively. Another important finding is that ILE238 plays an important role in maintaining the inactive state of the receptor. Two interaction interfaces were identified for the AT1R-AT2R heterodimer, a symmetric (TM3,5,6) and an asymmetric (TM1,2,8-TM3,4) one.
