Bacteria synthesize alarmones in response to stress conditions such as food deprivation and heat shock. Alarmones induce the accumulation of polyphosphate (polyP) polymer in the cell. PolyP in Esherichia coli is known to form a complex by binding to Lon protease, which is involved in many physiological processes that are vital to the cell, and this complex breaks down ribosomal proteins to form new amino acids for the synthesis of enzymes that provide adaptation to stress. It is not known whether there is such a relationship between poliP and Lon in other bacteria. Streptomyces coelicolor is used as a model organism for studies on the regulation of antibiotic production and these studies have shown that a diverse set of genes and external factors that cause stress affect secondary metobolite biosynthesis. In this study, we analyzed the transcription of genes involved in stringent response (relA), polyP metabolism (ppk), secondary metabolite production (actII-ORF4, redK) and stress response (sigB) in a high antibiotic producer recombinant S. coelicolor strain with an extra lon gene in its genome. We have seen a significant increase in the expression of these genes compared to wild type. Within the scope of this study, we have also created a mutant strain, (?ppk+pRAlon+Str), by integrating an extra copy of lon gene into the genome of high antibiotic producer S. coelicolor M145 ?ppk strain which cannot synthesize polyP. It is shown that this strain has slower growth rates compared to wild type and produce undetectable levels of antibiotic. We hypothesize that extra copy of lon gene generates high levels of stress in the absence of polyP, which in turn hinders vital activities. Results of this thesis not only provides the preliminary data that Lon protease-polyP interaction predicted in E. coli also exists in S. coelicolor but also sheds light on the complex network between Lon protease, (p)ppGpp, polyP and secondary metabolite production.
