White poplar (Populus alba L.) is a fast-growing deciduous tree species with high tolerance to abiotic and biotic stresses. This species is also a good candidate for afforestation studies, as it is resistant to changing climatic and environmental changes. Although white poplar generally reproduces sexually, it also reproduces vegetatively with root shoots. In fact, it is known that its clonal individuals are densely found in some areas. In addition, this species has been exposed to long-term human disturbance with the increasing population. Therefore, the genetic resources of the species are damaged due to natural biological events and anthropogenic effects. Hence, the thesis study aimed to evaluate the genetic diversity structure of white poplar populations in Turkey. In this context, leaves of 285 white poplar trees distributed in 15 different populations were collected, genomic DNA was isolated and isolated DNA was assessedusing 10 SSR molecular markers. As a result of the analyses, it was determined that the genetic diversity level of white poplar populations was relatively high (Ho: 0.50, He: 0.58). By AMOVA analysis, it was found that 84.20% of the genetic variation was within the population. In general, it was observed that as the distance between populations decreased, gene flow (Nm) increased and thus genetic differentiation decreased. PCA, structural and phylogenetic analysis data also supported these results. However, it was determined that the three populations located at the highest altitude andgeographically farthest (Erzurum-Oltu, Kayseri-Akkoy and Konya-Meram) were significantly different from the others. In addition to these three populations, it was observed that clonal reproduction is intense in populations close to the city center. It was found that white poplar populations sampled from riparian and floodplain forest ecosystems have higher genetic diversity and more unique alleles. In order not to lose this genetic diversity, it is recommended that the Eskisehir-Porsuk Stream, Edirne-Tunca River and Samsun-Bafra populations be protected in-situ. Our study provides information about the genetic diversity of white poplar populations in Turkey, thus contributing to more efficient and rational planning of programs aimed at breeding and conservation of the species.
