Ccdc124 (Coiled-coil domain containing 124) protein, which is conserved in all eukaryotes from yeast to human, is an RNA-binding protein and has been shown to be found in the ribosome and centrosome in the cell. Previous investigations have shown that overexpression and silencing of the CCDC124 gene in mammalian cell lines lead to cytokinesis being disrupted and the formation of multinucleated cells. However, it is still unclear how it affects cell division and other physiological processes taking place inside the cell. The aim of this study, which was carried out on the homolog of the CCDC124 gene in tomato plants, using the CRISPR/cas9, was to determine the morphological differences in plants by silencing the CCDC124 gene and to investigate its function. Additionally, this study is the first to examine the CCDC124 gene's plant homolog and indicate the phenotypic impacts of organism size in the absence of CCDC124 expression in plants, In the first step of this thesis, guide RNAs were designed to target the first exon of the CCDC124 gene, then cloned into the pYLCRISPR/Cas9P35S-N vector by Golden Gate cloning. Subsequently, Agrobacterium-mediated stable transformation of tomato plant was carried out. Sequence analysis was used to confirm mutations in the targeted region after PCR-based transgenic plant line identification. As a consequence of our research, it was determined that SlCCDC124 mutants had deletions of various lengths and base changes. Additionally, it was observed that SICCDC124 mutants have significant morphological differences compared to wild-type plants and premature senescence occurs in mutant plant lines.
