Özet

Streptomyces cinsi, uzamış ve dallanmış hiflerle karakterize edilen kendine özgü bir büyüme ve gelişim modeli sergileyen Gram-pozitif bakterilerden oluşur. En az beş farklı antibiyotik üreten Streptomyces coelicolor A3(2), genetik ve moleküler çalışmalar için yaygın olarak kullanılan bir model organizmadır. Protein homeostazında merkezi bir rol oynayan ATP-bağımlı Lon proteaz, birçok organizmada kapsamlı şekilde incelenmiş olmasına rağmen Streptomyces türlerindeki işlevi sınırlı sayıda çalışmada ele alınmıştır. Bu çalışmanın amacı, lon genini çok kopyalı bir plazmid üzerinde ifade eden rekombinant S. coelicolor A3(2) suşunda Lon proteazın büyüme, stres toleransı, antibiyotik üretimi ve gelişimsel düzenleme üzerindeki etkilerini araştırmaktır. Bunun için, lon geni gliserol ile indüklenebilir promotorun kontrolünde olacak şekilde pSPG vektörüne klonlanmış ve genin S. coelicolor hücrelerinde ekspresyonu RT-qPCR ile doğrulanmıştır. Rekombinant suş (Sco-Lon), 120 saatlik çentikli erlende yapılan fermantasyon sonunda yaban tipe kıyasla 27 kat daha fazla aktinorhodin ve 43 kat daha fazla undesilprodigiosin üretmiştir. Rekombinant suşun, morfolojik farklılaşması, sporulasyon verimliliği ve hücresel yapısı taramalı elektron mikroskobu (SEM), konfokal mikroskobu ve canlı spor sayımı ile değerlendirilmiştir. SEM analizlerinde, miselyum yapısı, spor zinciri organizasyonu ve genel morfoloji açısından rekombinant ve yaban tip suş arasında belirgin bir fark gözlenmemiştir. Sporulasyon oranları her iki suşta da benzer bulunmuştur. Buna karşın, canlı spor sayımı analizleri, Sco-Lon ve boş plasmid içeren kontrol suşu Sco-pSPG'ye ait sporların yaban tipe kıyasla canlılıklarında azalma olduğunu ortaya koymuştur. Konfokal mikroskop ile gerçekleştirilen zaman serisi analizleri, yüksek lon geni ifadesinin gelişimsel süreci belirgin biçimde etkilediğini ortaya koymuştur. Kontrol suşlarında Streptomyces'lerin hücre döngüsüne paralel şekilde hif gelişimi ve pellet oluşumu gözlenirken, Sco-Lon suşunda bu aşamaların düzensiz olduğu gözlenmiştir. Kontrol suşlarıyla karşılaştırıldığında recombinant pelletlerde çok daha yoğun programlı hücre ölümünün gerçekleştiği tespit edilmiştir. Rekombinant suşta 72. saatten itibaren canlı hücrelerin yalnızca pellet periferinde sınırlı kaldığı, ilerleyen zaman noktalarında ise pellet bütünlüğünün bozulduğu gözlenmiştir. Bunların yanında, lon geninin yüksek ekspresyonunun, rekombinant suşta ozmotik ve UV stresine toleransı anlamlı düzeyde arttırdığı bulunmuştur.Ayrıca, çalışma kapsamında, gelişimsel süreçlerde görev alan genlerin transkripsiyonel profilleri büyümenin 16., 48. ve 72. saatlerinde RT-qPCR ile analiz edilmiştir. Analiz edilen genlerin en önemlileri: havasal hiflerin oluşumundan sorumlu olan bld genleri (bldD, bldH, bldN, bldM), sporulasyonla ilgili olan whi genleri (whiA, whiG, whiH) ve sigF ile ssgB, hidrofobik kılıf oluşumundan sorumlu olan chpA and chpD genleri, divizom oluşumundan sorumlu ftsZ geni. Morfolojik farklılaşma ve gelişimden sorumlu olan genlerin ekspresyonunda tespit edilen değişiklikler, lon'un aşırı ekspresyonunun morfolojik farklılaşmanın zamanlamasını ve koordinasyonunu düzenleyebileceğini önermektedir. Genel olarak bu çalışma, Lon proteazın S. coelicolor A3(2)'de stres yanıtı, gelişimsel düzenleme ve ikincil metabolizmayı birbirine bağlayan kritik bir düzenleyici olduğunu göstermektedir. Elde edilen bulgular, Streptomyces'lerde ikincil metabolit sentezini etkileyen morfolojik, fizyolojik ve moleküler etkilere dair önemli bilgiler sunmakta ve endüstriyel açıdan değerli Streptomyces suşlarının geliştirilmesinde Lon merkezli genetik mühendisliği stratejilerinin potansiyelini vurgulamaktadır.

The genus Streptomyces comprises Gram-positive bacteria characterised by a distinctive growth and development pattern involving elongated, branched hyphae. Streptomyces coelicolor A3(2), a species that produces at least five different antibiotics, is a widely used model organism in genetic and molecular studies. While the ATP-dependent Lon protease plays a central role in protein homeostasis and has been extensively studied in many organisms, its function in Streptomyces species has only been addressed in a limited number of studies. The objective of this study is to examine the impacts of Lon protease on various physiological processes, including growth, stress tolerance, antibiotic production, and developmental regulation, in a recombinant S. coelicolor A3(2) strain that expresses the lon gene on a multi-copy plasmid. For this, the lon gene was cloned into the pSPG vector under the control of glycerol-inducible promoter, and its expression in S. coelicolor cells was confirmed by RT-qPCR. After 120 hours of fermentation carried out in baffled Erlenmeyer flasks, the recombinant strain produced 27 times more actinorhodin and 43 times more undecylprodigiosin than the wild type. The recombinant strain was evaluated in terms of morphological differentiation, sporulation efficiency and cellular structure using scanning electron microscopy (SEM), confocal microscopy and live spore counting. SEM analysis revealed no significant differences in mycelium structure, spore chain organization, or overall morphology between the recombinant and wild-type strains. Altough spore formation rates were similar in all strains, live spore counting revealed that spores from the Sco-Lon and the Sco-pSPG control strain, which contains an empty plasmid, exhibited reduced viability compared to the wild-type strain. Time-series analyses performed using confocal microscopy demonstrated that overexpression of lon gene significantly impacted the developmental processes. While control strains' hyphal development and pellet formation occurred in parallel with the cell cycle of the Streptomyces, these stages were irregular in the Sco-Lon strain. Compared to the control strains, much more intense programmed cell death was observed in the recombinant pellets. Live cells were confined to the periphery of the recombinant pellet at 72th hour of shake-flask fermentation, and the integrity of the pellet was disrupted at later time points. Additionally, overexpression of lon gene was found to significantly increase tolerance to osmotic and UV stress of recombinant strain. Within the scope of the study, the transcriptional profiles of genes involved in developmental processes were also analyzed by RT-qPCR at 16th, 48th, and 72nd hour of growth. The most important genes analyzed are the bld genes, which are responsible for forming aerial hyphae (bldD, bldH, bldN, and bldM); the whi genes (whiA, whiG, and whiH), which are together with sigF and ssgB genes, related to sporulation; the chpA and chpD genes, which are responsible for forming the hydrophobic coat; and the ftsZ gene, which is responsible for forming the divisome. Changes in the expression of genes responsible for morphological differentiation and development suggest that lon overexpression may regulate the timing and coordination of this process. Overall, this study demonstrates that the Lon protease plays a key role in linking the stress response, developmental regulation and secondary metabolism in S. coelicolor A3(2). These findings provide valuable insights into the morphological, physiological, and molecular effects that influence secondary metabolite synthesis in Streptomyces and emphasise the potential of Lon-centred genetic engineering strategies for developing industrially valuable Streptomyces strains.
