Liver cancer is the second leading cause of cancer deaths in men and the sixth leading cause of cancer deaths in women. Hepatocellular Carcinoma (HCC) is the most common type among the liver cancers. Viral hepatitis, aflatoxin intoxication and excessive alcohol consumption are the leading factors triggering HCC development. As in the case for other cancer types, genes and proteins involved in tumor progression must be thoroughly investigated in order for HCC to be diagnosed before the disease reaches end-stage fatal status. For this purpose, nine different HCC cell lines exhibiting distinct genotypic and phenotypic traits are used in this study. FLASH protein encoded by CASP8AP2 gene was shown to play roles in apoptosis, cell cycle and cellular proliferation. Thus, its expression is investigated in HCC cell lines and tissues by RT-qPCR, immunofluorescence and immunohistochemistry. Immunohistochemical analyses were performed in multiple tumor tissue arrays as well. Although there has been CASP8AP2 expression in all HCC cell lines, we didn't observed marked changes in expression levels. Gene expression levels were in accordance with the protein levels obtained by immunofluorescence microscopy. According to the immunohistochemistry studies performed on both various normal and cancer tissues, FLASH protein expression was only detected in healthy stomach tissue samples.
