HCC'de ROR1 ifadesinin incelenmesi tanı ve tedavi açısından önemli katkılar sağlayabilecektir. dc.description.abstractHepatocellular carcinoma (HCC) is among the most frequent and deadly cancers, yet tools for its early diagnosis and treatment are limited. The expression and role of receptor tyrosine kinase ROR1 in HCC have not been elucidated so far. Thus, the expression of ROR1 in HCC and its significance in hepatocarcinogenesis were investigated in this thesis work. Recombinant ROR1 protein was produced in bacteria and used as an immunogen for the generation of ROR1 monoclonal antibodies. The levels of ROR1 transcripts and proteins in HCC were detected by RT-qPCR and homemade monoclonal antibodies-based western blotting, respectively. The functions of ROR1 in hepatocarcinogenesis were explored through analyses of apoptosis, cell cycle, cellular migration and drug efflux in HCC cell lines, and cell clones in which the expression of ROR1 is repressed by shRNA. In addition, the expression of ROR1 was sought out in exosomes that were isolated from HCC cells lines and ROR1 dependent changes of miRNA cargo content of exosomes were analysed. The novel ROR1 monoclonal antibodies specifically recognized endogenous ROR1 protein both in human and mouse HCC cell lines. The suppression of ROR1 expression caused a decrease in cellular proliferation and cell migration, while the cells displayed increased resistance to apoptosis and anoikis. We also found that the chemoresistance of cells with repressed ROR1 expression is due to the increased drug efflux and upregulation of multidrug resistance genes. The expression of ROR1 was detected in HCC-derived exosomes and the expression of miRNAs that inhibit cell migration increased in the exosomes that isolated from ROR1-depleted HCC cells. ROR1 is expressed in HCC and contribute to the development of this cancer by affecting multiple signaling pathways. A detailed investigation of ROR1 in HCC will offer an insight into the diagnosis and therapy of HCC. This thesis supported by TUBITAK SBAG215S751 and 2214A Schoolarship.
