The human PD-1 protein was produced as a project output that can be used for cancer-related analytical purposes. PD-1 is one of the most popular protein that can able to inhibit the activation of T cells thus resulting in the proliferation of cancerous cells. Monoclonal antibody-based drugs which can block the development of cancerous cells by blocking the PD-1 protein, have taken their place on the market. In line with the given information, the PD-1 protein, which was expressed as a project output, was a potential final product that can be used in the bioassay development for monoclonal antibodies and therefore in cancer treatment studies. The mentioned thesis was based on optimization of expression and analytical potential of human PD-1 protein from the transiently transfected HEK293T (Human Embryonic Kidney) cell line. Cell viability after transfection was demonstrated by cell counter device, protein expression was checked according to GFP luminescence by using fluorescent microscopy, and quantified by using image analysis software. According to yield values, the data was investigated by using "design of experiment" tools to figure out the effects of parameters for higher expression rates. The cells expressed human PD-1 with optimized transfection parameters were used to screen the binding property of blocking antibody, by using a flow cytometry device and an ELISA test kit.
