Lon protease (also called La) is the first isolated ATP-dependent protease in E. coli. Lon is a member of the AAA+ super family (ATPase associated with diverse cellular activities) like all other ATP-dependent proteases (FtsH, ClpAP, ClpXP, HslVU ve 26S proteazom). Lon proteases are key enzymes responsible for intracellular proteolysis, contributing to protein control and cellular homeostasis by eliminating mutant and abnormal proteins and participation in rapid turnover of short-lives regulatory proteins. Homologs of the lon protease are found not only in bacteria also in human, yeast and plants. These observations indicate that Lon may play crucial biological roles in various organisms. However how Lon protease recognises a variety of substrates in ATP dependent manner is still unclear. In this study, Geobacillus kaustophilus ATP dependent Lon protease first time cloned and expressed in E. coli. About 2500 bp lon gene first cloned into pGEM-T cloning vector then subcloned into pGEX 6P-1 in E. coli. Lon, 87 kDa molecular weight, were expressed with glutathione S-transferaz (GST 26 kDa) as a fusion protein.
